Formaldehyde (FA) is an organic compound with the formula CH2O. In the medical field, it is frequently used in cadaveric anatomy and tissue fixation in histology and pathology. It is also used for disinfection processes in factories and various places. Formaldehyde, which is used as a preservative in the food industry with the code number E240, has negative health consequences.
It has been reported that epididymal sperm count and motility decreased in male subjects exposed to FA by inhalation and intraperitoneally. FA exposure is effective in the emergence of oxidative stress by reducing the levels of antioxidant enzymes. As a result, it has been observed that fertility is seriously affected by disruptions in spermatogenesis in testicular tissue and decreases in LH and testosterone hormone levels.
Tempol, a synthetic antioxidant, is a cell membrane-permeable nitroxide that directly scavenges free radicals resulting from oxidative stress.. In addition to its antioxidant effects, its antiapoptotic and anti-inflammatory effects have also been reported in studies. Tempol, which has protective properties against oxidative stress that occurs as a result of different diseases in organs such as kidney, brain tissue and lung, has limited effects on testicular tissue.

The aim of our study was to reveal the
possible effects of tempol in testicular tissue against FA exposure. Twenty
four male Wistar albino rats were divided into three groups as control, FA,
FA+Tempol. Every day for 14 days, saline will be administered
intraperitoneally to the control group, 10 mg/kg dose of FA to the FA group,
10 mg/kg dose of FA and 100 mg/kg dose of tempol to the FA+Tempol group. The
Nrf2/Keap1 signaling pathway, which regulates the transcription of
antioxidant stress genes, will be immunohistochemically evaluated from the
testicular tissues taken at the end of the experiment. In addition to this
signaling pathway, oxidative stress index and testosterone hormone levels
will also be determined to investigate the antioxidant activity of tempol and
its effect on sperm parameters. |
